An Experimental Model for Long-term Potentiation in Spinal Cord Produced by Electrical Stimuli Combined with Intrathecal KCI Injection in Adult Rat

Abstract
Although long-term potentiation (LTP) in spinal cord may be thought to contribute the mechanism of central sensitization in chronic pain, it is not easy to produce by tetanic electrical stimuli to peripheral nerve. In present study, we attempted to develop an experimental animal LTP model for chronic pain in adult rat. Under enflurane anesthesia the tetanic stimulation(group 1) or the burst-electrical stimulation to sciatic nerve of left side of rats (group 2) were conducted with the third group of rats (group 3) was treated burst electrical stimuli with intrathecal KCI. A parallel study was conducted with group 4, which received intrathecal KCI alone. Sensitivity of the hindpaw to mechanical stimulation was tested with von Frey filaments for the next 2 wk in all 4 groups. We did not observed any change of sensitivity for mechanical stimulation in the group 1. In group 2, however, threshold to mechanical stimulation was decreased from 15g to 8.5g and this behavior was maintained for one week. Group 3, in which excitation of spinal dorsal horn was induced by KCI treatment, shows that its threshold (3.6 g) was lower than group 2(8.5g) and maintained for 2 wk. In the group 4, we observed that threshold for left and right hindpaw decreased 1-2 d after KCI treatment, but this response disappeared after 3 d. Therefore, we believe that we have developed an experimental animal LTP model for using burst electrical stimuli combined with the intrathecal application of KCI. The present model suggests that input from primary afferent should be associated with depolarization of dorsal horn neurons to produce LTP.
